If you have a large group, or a group with diverse age group (especially if you have older folks, or very young kids), you could consider a group day tour such as this: http://bit.ly/3j8uano. If it's your first time, having a guide would also help you appreciate the history and cultural heritage of Ayutthaya better.

Day trip to Ayutthaya is easy. Doesn't require much planning to be honest. I've done it a number of times.

There are multiple trains from Bangkok to Ayutthaya every day. You can check the train schedule at the https://www.railway.co.th/Home/Index. Takes less than 2 hrs to get from Bangkok to Ayutthaya. The usual boarding points are Bangkok (Hua Lamphong) and the latest railway hub, Bang Sue Station. Both stations are linked /close to the MRT network.

Once in Ayutthaya, you can either rent a motorcycle, a bicycle or hire a local driver. Plenty of options that you can consider around the railway station.

Planning for my Jun BKK as well. Deciding between this package or the one for Maeklong + Amphawa Market. The former include Elephant camp... the later include Firefly watching... Both are from the same tour agency you have mentioned.
Looking at day tour for Ayutthaya as well as Death Railway as well.
Between Amphawa and Damnoen Saduak:
1. Amphawa is an evening / night floating market. Damnoen Saduak is all day round.
2. Amphawa is more popular with locals. Damnoen Saduak is more popular with tourists.
3. I was told that the prices in Amphawa is lower while Damnoen Saduak is slightly more expensive. However, cheap / expensive is really dependent on your bargaining skills (and personally, I do wonder whether it's worthwhile to spend 30mins haggling over THB50 ~ SGD2).
4. Both floating market are not that far from one another.

For the https://bit.ly/3X1fXav, I feel that the elephant camp isn't a must do. Hence, I chose the package without the elephant camp. Should you decide to purchase bananas to feed the elephants (which we did) or to experience an elephant ride in their camp / village (which we didn't bother with), you can do so on the spot.

Because of pt 1. above, it makes sense to include the firefly cruise for Amphawa. However, this would mean that you are going to get back to the city pretty late. For my day trip, I was able to get back to Bangkok (drop off either ICONSIAM or Siam Paragon) around 6pm. Plenty of time to either shop / eat or visit the other night markets.
